Nanodrop

Overview

We have a NanoDrop One/OneC UV-Vis (click here). This instrument offers full-spectrum microvolume UV-Vis measurements with a cuvette option in a single instrument. This instrument works on a dynamic range of 2.0–27,500 ng/µL for dsDNA, with lowest detection limit 0f 0.2 ng/µL with 10 mm cuvette.

<aside> ⚠️ The Thermo Scientific NanoDrop One/OneC Spectrophotometer is a compact, stand-alone UV-visible spectrophotometer for measurement of highly concentrated samples without the need for dilution across a wide variety of analytes. A glove-compatible touchscreen offers easy access to preloaded software applications and enables purity and concentration assessments in seconds. Built-in Thermo Scientific Acclaro Sample Intelligence technology helps identify and correct for the contaminants present.

Acclaro Sample Intelligence Technology

Thermo Scientific Acclaro Sample Intelligence Technology software (included with NanoDrop One/OneC and NanoDrop Eight spectrophotometers) analyzes absorbance across the whole UV-Vis spectrum, not just wavelengths specific to nucleic acids and proteins. It then applies powerful chemometric algorithms to find and correct for contamination in your samples. You get more reliable downstream assay results and avoid potential rework with this efficient, breakthrough capability.

Using the Nanodrop One

  1. Set up
    1. Sample diluent
    2. Labeled sample
    3. Pipette and pipette tips for 1-2 µL
    4. Kim wipes
  2. Select sample type
    1. Create a black using the sample diluent
    2. Wait for the instrument to conduct a self test and the follow instructions on the instrument’s screen
    3. Use a damped kim wipe to dab the lower and upper pedestals
    4. Apply 1-2µL of the diluent on the lower pedestal
    5. Press blank
    6. Repeat 2c–2d using your samples instead of the diluent; the Nanodrop instrument will begin reading your sample when it's arm is lowered
    7. Use the screen to end the experiment and export your data
    8. Clean the instrument one last time using a kim wipe with distilled water
